Porcine Pseudorabies Virus(PRV) gB or gE Antibody Rapid Test Kit 50

1. Principle
This test strip is based on colloidal gold immuno-chromatography
technique to detect porcine pseudorabies virus gB antibody in
porcine serum, plasma and whole blood qualitatively. When testing,
porcine pseudorabies virus gB antibody in sample combine with
antigen coated by colloid gold forming complexes, moving forward to
the other head. When reaching the T-line, the specific antigen on
the membrane capture the complexes and appear a T line, C line
appears, means the test is valid.

4. Sample preparation
4.1 Whole blood
Collect anticoagulated whole blood, use for that very day;
4.2 Serum
Collect blood, put at 37℃for 1-2h, take the supernatant, centrifuge
at 1500r/min for 10min,separate the serum. The serum can store for
2-3 days at 4℃, for long term storage, store at -20℃in frozen.
5. Operation procedures
1) Read the instructions carefully and return the test card and
sample to room temperature before use.
2) Take out the test card, draw the sample with a dropper, and drop
2-3 drops into the "S" hole; or use a micropipette to transfer 70ul
of the sample to the hole marked "S".
3) Read the results in 15 minutes at room temperature.
6. Results
Negative(-): no color reaction on Test Line (T Line),only on
Control Line(C Line) being seen red color reaction. It means there
is no porcine pseudorabies virus gB antibody in sample or the
levels is lower than the detection threshold;
Positive(+): Both on T Line and C line being seen red color
reaction. It means there is porcine pseudorabies virus gB antibody
in sample;
Invalid: No color reaction on C Line. It means the operation is
wrong or the test card is invalid.
7. Notice
1) The test card can only be used once at room temperature, please
do not reuse or use expired test cards.
2) Bring all reagents and samples to room temperature before use.
3) Be careful when preparing samples, wear gloves and a mask.
4) Whole blood needs to be added with anticoagulant and cannot be
frozen; serum samples can be frozen to avoid repeated freezing and
thawing.
5) This product is only for rapid veterinary in vitro diagnosis,
and cannot be quantified, but only qualitatively tested.
6) Product test results are for reference only, please refer to
relevant national standard methods for confirmation.
Storage and expiry date
Storage: Store at 4-30℃ in dark, sealed, dry place, no frozen.
Expiry date: 12 months; date of production is on box.
Clinical symptoms of porcine pseudorabies virus
The clinical manifestations of pseudorabies virus mainly depend on
the virulence and amount of infection of the virus, as well as the
age of infected pigs. Among them, the age of infected pigs is the
most important. Like herpesviruses of other animals, young pigs are
the most ill after being infected with pseudorabies virus.
Newborn piglets infected with pseudorabies virus will cause a large
number of deaths. Newborn piglets will behave normally on the first
day of clinical diagnosis, and become ill from the second day. The
peak period of death is 3 to 5 days, and some whole litters will
die. At the same time, the infected piglets showed obvious
neurological symptoms, lethargy, whining, vomiting, diarrhea, and
died within 1 to 2 days once the disease occurred. At autopsy, the
kidneys were mainly covered with needle like bleeding points,
sometimes pulmonary edema, meningeal surface congestion and
bleeding were seen. Piglets less than 15 days old infected with the
disease are very serious, and the morbidity and mortality rate can
reach 100%. Piglets suddenly get sick, their body temperature rises
to more than 41 ℃, they are extremely tired, shivering,
uncoordinated movements, convulsions, vomiting, diarrhea, and
rarely recover. Weaned piglets infected with pseudorabies virus
have a incidence rate of about 20%~40% and a mortality rate of
about 10%~20%. The main manifestations are neurological symptoms,
diarrhea, vomiting, etc. Adult pigs are generally recessive
infection, and the symptoms are mild and easy to recover. The main
symptoms are fever and depression. Some sick pigs vomit and cough,
and generally recover completely within 4 to 8 days. Pregnant sows
may miscarry, give birth to mummified fetuses or stillbirths, of
which stillbirths are mainly seen in first born sows or multiparous
sows, and there is no strict seasonal pattern, but they often occur
in cold seasons, i.e. late winter and early spring.
Another characteristic of pseudorabies is the sterility of breeding
pigs. Sows are sterile after repeated mating, and the rate of
return to estrus is as high as 90%. Some sows can not be mated
after repeated mating for several times. In addition, boars
infected with pseudorabies virus showed swollen and atrophic testes
and lost the ability of breeding.
