Labnovation SPERM CONCENTRATION TEST is a rapid test for qualitative detection of sperm concentration in human semen, as an aid in the determination of a man’s fertility status. It is suitable for in vitro qualitative detection of sperm SP10 protein in human semen. The WHO "Human Semen Testing and Processing Laboratory Manual" (Fifth Edition) mentioned that the main factors affecting male infertility are: Semen volume, total number of sperm, sperm concentration, liquefaction time, semen pH value, sperm morphology rate, etc., and sperm concentration detection is an important indicator of male infertility. In this manual, the normal minimum value of sperm concentration is 15x106/mL. When the sperm concentration is less than 15x106/mL, it is considered that there is difficulty in fertility. But male infertility needs to be combined with other indicators to make the final judgment. Based on several anatomical, genetic and biochemical characteristics. SP10 protein was confirmed to be an analyte for sperm concentration detection. According to research data, there is a certain linear relationship between SP10 protein concentration and sperm concentration, when the sperm concentration is> 15x106/mL, the SP10 protein test is positive. Therefore, the sperm concentration can be used for the auxiliary diagnosis and curative effect observation of male infertility, and to provide guidance for the birth plan of the right-age couples.

TEST PRINCIPLE

 

This kit is an immunochromatography assay and uses double antibody sandwich method to detect the specific protein SP10 in human semen. During detection, when the concentration of SP10 protein in the semen sample is higher than or equal to the minimum detection limit, the sperm SP10 protein in the semen combines with the SP10 monoclonal antibody labeled with colloidal gold to form a reaction complex, which is reacted by chromatography moving forward along the nitrocellulose membrane, it is captured by the SP10 monoclonal antibody pre-coated on the detection zone (T) of the nitrocellulose membrane, and finally a red reaction line visible to the naked eye is formed. At this time, the result is positive; on the contrary, When the semen sample does not contain sperm SP10 protein or the sperm SP10 protein concentration is below the minimum detection limit, no red reaction line appears in the detection area (T), and the result is negative at this time. No matter whether the sample contains sperm SP10 protein; in quality The control area (C) will form a red reaction line.

PROCEDURES

1. Collect the semen sample into the semen collection cup. If you do lose some of the semen, discard the sample. Rinse the cup ONLY with tap water. Wait at least 48 hours, but not more than 7 days (see 1 above.) and collect a fresh sample.
2. Let semen stand for at least 20 minutes but not longer than 3 hours at a room temperature (15-30°C) upright on a flat surface, until the sample has visibly liquefied.
3. Gently stir the semen sample with the semen transfer syringe in the cup about 10 times until it is well mixed.
4. Insert the semen transfer syringe into the semen sample avoiding any solid or sticky material within the semen. Pull the plunger to draw the sample into the semen transfer syringe until it reaches the 0.1ml line on the syringe. Avoid getting air bubbles in the semen transfer syringe. If this happens, push the semen back out completely and then draw semen into the semen transfer syringe again.
5. Solution bottle has two caps. Open the solution bottle by unscrewing the entire cap.
6. Insert the semen transfer syringe into the solution bottle and push the plunger to add the entire semen sample (0,1ml) to the solution bottle.
7. Screw the cap back and mix the con- tent of the solution bottle by turning the solution bottle gently upside down. Do not shake the solution bottle as this could cause foaming.
8. Let the solution bottle stand for 2 minutes to create the semen mixture. If this does not happen, turn the solution bottle upside down again. Open the foil pouch and remove the test cassette. Lay it face up on a clean, dry and flat surface.
9. Twist off the small cap on the tip of the solution bottle cap. Hold the bottle with the semen mixture straight up and down over the test cassette and squeeze gently to add exactly 3 drops to the sample well (S). Please note, that there should be no liquid ap- plied to the result windows marked with the letters (T) and (C).
