Porcine Parvovirus (PPV) Antibody Rapid Test Kit 50 Tests/Kit GMP

 

1. Principle


This rapid test is an immuno-chromatographic test, using colloidal gold immunoassay method, to detect the antibody against porcine Parvovirus in whole blood, serum. After adding sample, the sample move along with the colloidal gold labeling PPV antigen, if there is PPV antibody in the sample, it combines with antigen on T line and show wine-red color; if there is no PPV antibody, then no color reaction.

3. Test procedure

 

1) Collect whole blood, separate serum (centrifuge at 2000-3000 r/min for 5~15min or 4°C overnight for natural separation), or take whole blood containing anticoagulant as a sample. Whole blood without anticoagulant can also be used as a sample, but it must be tested immediately.
2) Open the package, take out the test card, and place it on a clean and flat table.
3) Aspirate the sample with a dropper and slowly drop 3 drops of the sample into the "S" marked hole.
4) 10-20min judgment result, more than 20min is invalid.


4. Results judgement


1) Positive: control line and test line are both seen wine red, the more the antibody exist, the thicker the color appear;

 

 

2) Weak positive: control line and test line are both seen wine red, but the color of test line is very light;

3) Negative:only control line is seen wine red;

4) Invalidation: control line isn’t seen wine red.

 

5. Consults for diagnosis


1) If not inoculated by the bacterin against porcine Parvovirus:
a. test line don’t appear visible wine red show there are no antibodies against porcine Parvovirus virus in tested samples, if the swine are healthy, it is better to vaccinate in time;Parvovirus infection cannot be ruled out if the corresponding acute symptoms occur.
b. test line appear visible wine red show the pig is present with parvovirus wild-type infection or previous infection, should be more observed to validate.


2) if have been inoculated by the bacterin against porcine Parvovirus:
a. the color of test line is thicker than the color of the titer 1:40 from comparison card show the high titer of the antibody against porcine Parvovirus, which is enough to resist the strong virus, meanwhile, it is less likely that pigs will be infected with PPV;
b. the color of test line is thinner than the color of the titer 1:40 from comparison card show the the titer of the antibody against porcine Parvovirus is not enough to resist the strong virus, there is a risk of wild virus infection, it is better to perform complementary vaccination.


6. Storage and expiry date


Store at 2 - 30ºC and the shelf life is 24 months.


7. Notice

 

1) Do not use expired or poorly packaged test cards;
2) Do not use purified water, tap water and normal saline as negative controls;
3) Store at room temperature (< 30°C). If it is stored in a cool place, it is necessary to return to normal temperature first, and then open the package to avoid moisture. Use the test card as soon as possible after opening the package.
4) Do not touch the white film surface in the middle of the test card.
5) To ensure the quality of the test, please take fresh, non-contaminated samples. If there is a lot of blood lipids, it will affect the results, so after centrifugation, take the serum test to avoid fat. If the blood or serum is viscous or not viscous enough, it can be diluted with normal saline at the same dilution and then tested.
6) If the serum cannot be detected immediately, it can be stored at 4°C for a short time (within 48 hours) and for a long time at -20°C.
7) Whole blood without anticoagulant should be tested immediately; if some Krull is already present, separate serum for testing. Whole blood containing anticoagulants should be tested within 24 hours and not frozen. Freezing and thawing lead to hemolysis and rupture of red blood cells, which seriously affects the test results.
8) When adding the sample dropwise, keep the pipette tip and the "S" hole 1cm to ensure accurate sample addition. If it is too close, it will cause the droplet volume to be too small, resulting in wrong judgment of the droplet or not allowing the amount of sample to be added, which will affect the test results.
9) Avoid foaming when adding samples, and the sample volume should be controlled at about 100ul (about 3 drops). Too many samples will not increase sensitivity. In particular, excess whole blood causes erythrocytes to settle in the "well", causing membrane chromatography to penetrate deep into the red background, which seriously affects the results.
10) Antibody level is only related to T-line darkness, not C-line. Only the C wire is clean, indicating that the test is valid.
11) Dispose of the test cards and samples at the contaminated site according to local regulations.

 

Epidemiology of Porcine Parvovirus


Domestic pigs and wild pigs of different ages and genders are susceptible. The source of infection mainly comes from sows infected with parvovirus and infected boars. The gilts are more susceptible to infection than the sows after childbirth. The virus can be transmitted vertically through the placenta, while the live pigs produced by infected pigs may be infected for a long time or even for life. The infected boar is also the most dangerous source of infection of the disease. The virus can be isolated from the boar's semen, spermatic cord, epididymis and gonad. The boar can be transmitted to susceptible sows through breeding and spread the disease.
