INTENDED USE
 
Dog Relaxin (RLN) Rapid Test Cassette (Serum/Plasma) is a sandwich lateral flow immunochromatographic assay for the
qualitative detection of canine Relaxin (RLN) in dog’s serum and plasma 15 days after the last mating of the dog, for the auxiliary diagnosis of early pregnancy.
 
BACKGROUND INFORMATION
 
Relaxin is a peptide hormone synthesized and secreted by corpus luteum and placenta during pregnancy. The secretion of relaxin in pregnancy increased gradually with the increase of gestation and decreased rapidly to undetectable after partum or
spontaneous abortion. The serum relaxin level of pregnant dogs at 15 to 30 days is significantly higher than that of not pregnant dogs, peaking at 35 days, so relaxin can be used as an early pregnancy marker in dogs.
 
PRINCIPLE
 
Dog Relaxin (RLN) Rapid Test Cassette is based on sandwich lateral flow immunochromatographic assay. The test cassette has a testing window. The testing window has an invisible T (test) zone and C (control) zone. When sample is applied into the sample well on the cassette, the liquid will laterally flow on the surface of the test strip. If there is enough dog relaxin in the sample, a visible T line will appear. The C line should always appear after a sample is applied, indicating a valid result. By this means, the cassette can accurately indicate the presence of canine relaxin in the sample.
 
STORAGE AND STABILITY
 
Store as packaged in the sealed pouch either at room temperature or refrigerated (2-30 °C). The test is stable through the expiration date printed on the sealed pouch. The test cassette must remain in the sealed pouch until use. DO NOT FREEZE. Do not use beyond the expiration date.
 
DIRECTIONS FOR USE
 
Allow the test cassette, specimen, buffer, and/or controls to equilibrate to room temperature (15-30 °C) prior to testing.
 
1. Venous blood was collected and centrifuged at 2000-3000rpm for 5-10 minutes to obtain dog’s serum or plasma. Only use clear, non-hemolyzed specimens. Serum and plasma specimens may be stored at 2-8 °C for up to 3 days. For long term storage, Serum and plasma specimens should be kept below -20°C.
 
2. The serum or plasma should be diluted 1:1 with buffer provided. For example, you can add 3 drops of buffer to the extraction tube, then transfer 3 drops of serum or plasma sample into the extraction tube with dropper provided. Press the bottom of the extraction tubes to mix the sample thoroughly, then fit on the extraction tube tip onto the plastic tube and place it on the workstation for standby.
 
3. Place the test cassette on a clean and level surface.
 
4. Inverted the extraction tubes and add 2 drops (approx. 80μl) of extracted specimen to the specimen well on the test cassette. Please avoid trapping air bubbles in the specimen well and do not drop any solution in observation window.
 
5. Wait for the colored line(s) to appear. The result should be read at 5 minutes, do not interpret the results after 10 minutes
 
Positive: The presence of both C line and T line, no matter T line is clear or vague.
Negative: Only clear C line appears.
Invalid: No colored line appears in C zone, no matter whether T line appears.
 
SENSITIZDE AND SPECIFICITY
Dog Relaxin (RLN) Rapid Test Cassette has been evaluated with specimens obtained from veterinary hospital. B-type ultrasonic is used as the reference method for the RLN Test. Specimens were considered positive if B-type ultrasonic indicated a positive result.
